KRAS Signaling Inhibition Induces a Targetable Metabolic Dependency on Lipophagy-Dependent Fatty Acid Oxidation in Pancreatic Cancer

1 month 4 weeks ago
Pancreatic ductal adenocarcinoma (PDAC) is characterized by frequent KRAS mutations, which activate the MAPK pathway to promote PDAC progression. Here, we explored metabolic vulnerabilities of PDAC by assessing initial metabolic reprogramming upon ERK inhibition using metabolomics, lipidomics, and isotope-tracing experiments. ERK inhibition enhanced lipid turnover and fatty acid oxidation while inhibiting glycolysis, glucose oxidation, and glutamine metabolism in PDAC cells. Moreover, lipophagy,...

SGLT2 inhibitor dapagliflozin attenuates HIV-associated cardiac fibrosis, steatosis and diastolic dysfunction in a mouse model via inhibition of TGFβ signaling

1 month 4 weeks ago
Myocardial fibrosis, steatosis, and heart failure with preserved ejection fraction are increasing among people with HIV (PWH). The sodium-glucose cotransporter type 2 inhibitor (SGLT2i) dapagliflozin has efficacy for cardiovascular disease (CVD) prevention in type 2 diabetes and is a promising therapy for the inflammatory component of CVD risk in PWH. We show dapagliflozin preserved diastolic function and suppressed cardiac fibrosis and steatosis linked to HIV in mice, suggesting potential...

Co-Curricular Science Experiences Support Undergraduate Students' Science GPA, Degree Completion, Workforce Entry, and Motivation

1 month 4 weeks ago
Science co-curricular experiences (SciXP)-such as research experiences and internships-are a common, yet understudied, strategy to enhance undergraduates' science career pursuit. Using a doubly robust method to account for initial differences in science motivation, academic readiness, and career intentions, we investigated whether SciXP participation across four years of college predicted undergraduates' (N = 1984) science achievement, career persistence, and motivation at the end of college and...

Deep profiling of lupus nephritis kidneys reveals dynamic changes in myeloid cells associated with disease progression

1 month 4 weeks ago
CONCLUSIONS: We suggest a detailed picture of the changes in the kidney immune mechanisms in LN as this disease progresses.
